Manufacturing of gene therapy vectors remains a complex process requiring close monitoring of viral vector titer and quality during production and bioprocessing. ELISA has been the gold standard for immunoassays measuring lentiviral and AAV titers, however this assay format is time- and sample-consuming.
This case study demonstrates the transfer of the Progen AAV Titration ELISA assay to Gyrolab immunoassay system for the characterization of AAV manufacturing preparations. In comparison to manual ELISA, Gyrolab immunoassay provides better sensitivity and dynamic range, and can quickly analyze 10 μL of sample. Gyrolab nanoliter-scale immunoassays produce meaningful data within 1 hour to make time-critical decisions during the development and production of gene therapeutics.
